Annexin V-fITC kit allows fluorescent detection of annexin V bound to apoptotic cells and quantitative determination by flow cytometry. The AnnexinV-fITC kit uses annexin V conjugated with fluorescein isothiocyante (fITC) to label phosphatidylserine sites on the membrane surface. The kit includes propidium iodide (PI) to label the cellular DNA in necrotic cells where the cell membrane has been totally compromised. This combination allows the differentiation among early apoptotic cells (annexin V positive PI negative) necrotic cells (annexin V positive PI positive) and viable cells (annexin V negative PI negative).

Annexin V is a cellular protein, which plays important roles in the inhibition of the activity of phospholipase A1 and blood coagulation by competing for phosphatidylserine (PS) binding sites with prothrombin. Annexin V has a high affinity to PS and is used as a probe to detect cells that have expressed PS on the cell surface. After initiating apoptosis, cells translocate phosphatidylserine (PS) from the inner face of the plasma membrane to the cell surface soon, which can be easily detected by Annexin V. Annexin V is a member of the annexin family of proteins that bind to membrane phospholipids in the presence of calcium. This dye has high affinity for phosphatidylserine (PS) that is present in the inner leaflet of the plasma membrane. Verified for use with flow cytometry assays use this antibody to detect the translocation of PS to the external environment. Annexin V is used along with viability dyes such as 7-AAD (Catalog 75001) or Propidium Iodide (Catalog 75002). The process of PS translocation occurs prior to the loss of membrane integrity. Therefore as cells progress through apoptosis and towards necrosis the cell membrane is compromised and consequently viability dyes pass into the cell. Thus cells undergoing early apoptosis stain positive for Annexin V and negative for viability dyes while apoptotic death or necrosis is characterized by positive staining for both Annexin V and the viability d
